Knilonat 200 mg (Nilotinib)
Knilonat 200 mg contains nilotinib, a second-generation BCR-ABL tyrosine kinase inhibitor (TKI) used for the treatment of Philadelphia chromosome–positive chronic myeloid leukemia (Ph+ CML). It is more potent and selective than imatinib and is used in chronic phase and accelerated phase CML where molecular response is required.

- Standard dose: 300 mg twice daily for newly diagnosed chronic-phase CML (using 200 mg capsules appropriately).
- Alternatively: 400 mg twice daily for imatinib-resistant/intolerant patients.
- Take doses approximately 12 hours apart.
- Must be taken on an empty stomach — avoid food for 2 hours before and 1 hour after each dose.
- Swallow capsules whole with water.
- Do not skip doses or stop treatment unless your hematologist instructs.
Important: Taking nilotinib with food dramatically increases drug levels and can cause dangerous heart rhythm abnormalities.

Nilotinib binds to and inhibits the ATP-binding pocket of the BCR-ABL oncoprotein, preventing phosphorylation and downstream signaling that drives leukemic cell growth. This restores normal myeloid cell maturation and reduces abnormal proliferation.
